The fire broke out in the early hours above a cafe called Dish in Bridge Road, East Molesey and flames and smoke could be seen above the building.
Firefighters used an aerial ladder to douse the flames across the roof and water was pumped from the Thames to support the water supply.
It is understood two casualties suffering from smoke inhalation were taken to hospital.
